Would it be possible to add a sort of Religious victory to the main game? Like, maybe 75% of the world population is of your state religion? (And you'd have to control the holy city for that religion.)

Look at "Assets/XML/GameInfo/CIV4VictoryInfo.xml". You can add a religious victory.
To verify if the winner has the Holy city, I suppose that you'll have to add some python...
