If I create multiple new buildings in a BTS mod, and all of these new buildings use the same ART DEF (from a building originally included in the game), could this possibly cause the mod to CTD? I'm suspecting it does, but I'm not certain. Does anyone know about this by any chance?

It shouldn't. Try it, but I'm sure duplicate or triplicate buildings isn't an issue since most early mods had to re-use vanilla graphics.

Well, I have tried this. The mod works for 30-40 turns, rather slowly, and then CTD's during the AI's turn. I noticed that a game in progress cannot be saved either, including the auto saves (I just get a zero k file). The good news is -- it's not the buildings. Now I have no clue what the problem is... :(

Found the issue -- needed to remove an unwanted comma from the button tag in one of the buildings' art def entry. Problem fixed.
