I know you can't make a unit gov specific
But I think you can make an improvement/wonder gov specific (is this true?)
I've heard these effects stop if you change gov (true?)
Can you make a unit require this wonder to build?

Not currently. You can't make a building (regular, small wonder, or big wonder) be a prereq for a unit. Maybe this fall you will be able to :p
Edit: And Yes, you can make buildings available only to certain gov'ts and I believe now the bug has been fixed so that the effects of that building do only apply during that gov't

Manhattan Project has an "Allows Construction of Nuclear Devices" flag set on the Wonders and Small Wonders section of the Improvements/wonders tab. ICBMs and tactical nukes have a "Nuclear Weapon" flag set in the Unit Abilities section of the units tab. This is an extreme special case. I suppose one could mess with these in a very limited context for a scenario, allowing a single small wonder for each civ allow those civs to build a special unit or group of units, perhaps when only in a certain government.(*) However since this is a single flag combo, this would remove the Manhattan project as nuke prereq ability and it is nowhere near the kind of generic solution a simple "Unit X requires building Y" setting would be.
Edit: (*)Actually, even that might not work, because I think the Manhattan flag is simply flipped 'on' upon its construction and then never flipped back 'off.'

No, not quite, there's still problems with a few of the flags for wonders. It looks like all the small wonder flags still operate even if you swich govs.
