Here are some challenges that might add more flavor to the game. I’m planning on adding more later if there’s positive feedback. Please add your ideas.

An easy one first.
1. Prom Queen
Player civ: any of the following: the Celts, Russia, Carthage, England, Spain, Portugal, Austria, Byzantium, China
AI civs: any of the above
Victory type: Diplomatic
Special rules: All civilizations must be in control of their original capital (it’s just a prom). You must liberate conquered capitals.

2. Those are my toys!
Player civ: any of the following: Mongolia, Venice, Austria
AI civs: the other two from above. Add civs that benefit CSs in some way: Greece, Sweden, Siam.
Victory type: any
Special rules: The player must be in control of all CSs at the end of the game (annex/puppet/ acquire by diplomatic marriage or great merchant of Venice).

3. Pedro and the wolves
Player civ: Brazil
AI civs: any of the following: Assyria, Songhai, the Huns, Carthage, Mongolia, Sweden, Aztec, France, Japan, the Zulu. Add someone else, if you’d like to play on a huge map.
Victory type: Cultural
Special rules: The player must not declare war or annex/puppet cities (may liberate them though).
 
Religious Domination:

Player Civ: any (religious recommended)
AI civs: all other religious ones + extras if on huge

Special Rules:
Tier 1: must have all cities worldwide following your religion
Tier 2: must wipe out the holy city status in all other world religions with an inquisitor so they can never come back

Victory Type: Any, but you can't do 2 without domination really

I'm getting close on this huge game but not playing domination so I'll never get tier 2 (see below) My goal is to unify the world under Zoroastrianism!

On this particular immortal, small islands game 3 other civs went fullblown piety so if it can be done here I think it's valid on most setting/maps/difficulties.

Here's two:

1. Irrational citizens : You may not ever open the Rationalism tree.
In addition, if any AIs actually take Rationalism you must punish them by conquer their capital(s) before winning. Other than that you can win any way you want.

2. Honor before reason: You must open and complete the Honor tree before anything else. Map size must be at least standard size and you may not reduce number of AIs below normal for the map size either.
In addition, you must conquer the capitals of all dishonorable AIs (those that didn't take at least one policy within the honor tree) before winning but can win any way you want.
 
Here's two:

Let's expand it!

3. Proletariat Revolution

Play as Russia/Soviet Union (mod). Before industrial era stay rather calm and isolated, although normal participant of world diplomacy. Get Order and attack all civilisations which don't follow Order. :D
Make the entire world follow only Order either by conquest of Free/Autocratic capitals or by... well... revolutions due to your massive tourism.


4. Nazi Insanity

Play as Germany/Nazi Germany (mod). Play with America, Russia, England, France, Poland, Rome, Japan, whatever else. Before modern era stay rather peaceful and play TALL, be developed but tall. Get Autocracy and declare war on:

- all Order and Freedom civilisations
- America, Russia, England, France, Poland
- all civilisations which are not Rome/Japan
- ...and get all their capitals and city states :D

5. Murica!

Play as America :D Set whatever opponent civilisations but among them has to be Russia and Arabia. Stay wide, powerful but 100% pacifistic until atomic era. Get Freedom. In atomic era declare war to Russia, Arabia and all Order/Autocracy civilisations at once.
Control Mecca, Moscow and all their capitals.
BONUS: have 100 oil supplies :D (including used by your units)

6. Colonisation

Play as... Well, whoever, but England would be particularly sexy. Continents map. Go tall and you can't conquer anything on your own continent. Since renaissance era you can start expansion but only overseas. Conquer half of overseas civilisations (including all their cities) before atomic era.

7. Voice of Prophet

Play as Arabia. Stay calm and isolated until medieval era but follow Piety and develop Islam religion with following beliefs: Desert Folklore, Pilgrimage, Holy Warriors, Mosques, Just War, Religious Fervor. In medieval era get third great prophet and immediately start world conquest against all civilisations which don't follow Muslim religion. By the end of game you have to either win domination victory or convert all world capitals on Islam.

8. 2012

Play as Maya. Stay scientific and religious superpower but rather peaceful. Be developed but peaceful for the entire game... Until information era. Don't allow anyone on winning the game.

In 2012 AD turn you have to nuke all world capitals simultaneously and capture them :D
