The forces of Charles V entered Rome in 1527 and famously sacked it, keeping Pope Clement VII a virtual prisoner in the Castel Sant'Angelo. The ramifications of this were quite extensive, but in this case, Spain did indeed conquer Rome (however unintentionally).
