Using World Builder I added a second settler at the start up. The second settler doesn't have the ability to settle. It's not even an option given. Is there a certain distance they have to be apart from each other? Or am I missing something?

They do have to be a few squares away, but you'd get a message telling you that.
If you used the World Builder:
1 - be sure you gave the Settler to you and not to a competitor
2 - be sure you've closed World Building (...it happens)

There is a certain distance, but if you are within that distance then it will show the option but make it greyed out and unclickable. If you hovor the mouse over the "Build city" button you will see it will say "Must not be within ___ plots of a city".
Sure you dont have One City Challenge setting turned on?
