1. I have Uranium, a mine and a road, but still tells me that I have 0 Uramium. What's the problem here?

2. What's the best way to defend an Offshore Plataform?

1) Difficult to tell, but it might be that you have a trading deal in which you are selling it to someone. Or you are paying a ransom to someone with it. If that's the case, you should cancel the deal.

2) I think it's keeping your battleships/missile cruisers (if I remember their name well) on it or maybe next tile to it (but it won't safe it from missiles then) with "patrol" mission.

3) Yes, but you require plastics if I remember well.

Whilst Physics reveals Uranium, you actually need Fission to be able to mine the Uranium. Building a mine or a fort on the resource won't do anything until you discover Fission, but it is a good idea to build them before that so that you instantly get the resource upon discovering Fission (in a similar way to how you'd build a fort on some incense before discovering calendar).

As Munch said, you need fission as well to use the uranium.

Station a fleet of battleships on it, especially if this is your only source.

Build a workboat and tell it to do so; but only after you have researched plastics.

One of my recent offline games I found myself without land-based oil and really couldn't expand/conquer to get some so instead I found uranium and tried mining it. Doh, still ain't got none, so I beeline fission to enable the uranium and then work toward plastics to claim the offshore oil that I do have access to while building uranium powered battle ships to guard it.

Destroy everyone who might threaten it. :)

Seriously speaking, battleships are good choice and have atleast one medic promotion in stack because enemy battleships causes collateral damage. Station some fighters to nearby cities and give intercept orders for them to prevent enemy airforces from bombing your platform.

There is still one trick that I like to use to destroy AIs oil rigs. A submarine loaded with cruise missiles. But I haven't seen AI using this tactic. Does anyone know is AI smart enough to destroy oil rigs or fishing boats with cruise missiles?
 
2. Place some destroyers/battleships on it, but if the potential for a hostile fleet to blow them out of the water is high, keep a good reserve in port nearby, along with a spare workboat so you can (1) wipe out the enemy fleet, and (2) to restore your oil supply.
 
Oil is a "must have" resource at the start of the modern era, and if you don't have any, or it is located in a bad location, you may need to change your overall strategy slightly. The AI civs don't like to trade Oil on the higher difficulty levels, and often don't have any spare Oil to trade anyway. As such, it's unwise to rely upon foreign trade to supply you with Oil, and if you do, then you are wide open to the AI cutting it off at just the wrong moment.

The two worst locations for Oil are right on the edge of your borders with a neighbouring civ, or offshore.

You need a large and mixed naval force to defend an offshore platform, and battleships alone are inadequate as they can't see or attack enemy subs, so you need destroyers (which can see / fight subs), plus battleships, (which can fight off enemy surface ships). You also need nearby fighter cover, and a few cruise missiles and/or bombers nearby in case you have to soften up a huge stack of enemy naval units.

You never know in advance if your civ has any Oil, or where it's located, so it's a good insurance policy to plan on it being offshore. Locating national wonders like Heroic Epic / West Point / Iron Works in your most productive coastal city give you an insurance policy, as does settling a few Great Generals in that city along with a Military Academy.

You will then be ready to quickly churn out strong naval units that are already promoted, and will be able to defend an Offshore Platform with a smaller naval force, that won't take as many casualties.

If you find yourself in a situation where you have no oil, or it is located offshore and you are not in a position to defend your Offshore Platform, then like I said you may need a slight change of strategy towards the end game. I have played loads of games whereby I planned to avoid war completely, because my end game strategy was based upon a peaceful victory via space / culture / diplomacy. Such plans often ended up in the trash bin upon discovering I didn't have any Oil, or it was offshore, and bloody difficult to defend because my coastal cities were very non-productive. Such scenarios usually ended up with me having to go to war to get the safe supply of Oil that I needed.

These days I make sure that I always found or conquer at least one highly productive coastal city, as that one city can easily defend an offshore platform if you feed it the items I mentioned.

2)Stick a few battleships or missle cruisers on the tile. To be really thorough, add a couple of carriers of fighters and a medic promoted destroyer. If it really is that important (i.e. it's your only source of oil), try and station ships so you can see incoming enemies, and attack them before they reach the platform.

3)You need a workboat, and the plastics tech.
