Hopefully someone can assist with your exact issue but assuming you still have your old machine and a flashdrive, you could just copy over your entire civ folder. There's no need to install it/patch it/etc. unless of course you're seeking a fresh install for whatever reason. Do you still have your old machine and can you just copy and paste the game over?
 
All I can suggest is re-booting your computer. That should close all running programs. Unfortunately, I haven't used Windows regularly for a few years, so I can't offer much advice beyond that.

Tech, as you you have installed Civ 2 ToT successsfully on your Win 10 pc and only the update of the patched Civ 2 ToT exe cannot be done, it would be sufficiant to copy your old patched Civ 2 ToT exe from your old laptop installation into the current Civ 2 ToT installation of your Win 10 pc and to overwrite the unpatched exe with the old one from your laptop.

I had similar problems with my Civ 2 ToT Chronicles installation, that were solved by this procedure.
 
Tech, I´m not a computer specialist. As far as I remember, this is not a substitute for the official patch, but it overcomes problems for 64 bit machines. The first of the ToTPP patches also holds a fix for 64 bit machines, too:

For starts of Civ2 ToT without ToTPP on a Win 10, 64 bit machine, I think you need that program to run the game. I am convinced there are many civers here in this forum, who can explain this much better than me, but as I was asked, of course I try to answer it as good as I can. :)

I've got a new computer with Win 10. I installed ToT with no problem, but it won't let me install the (official) patch. The popup says I have to close uninstallShield first, but I can't find it. I opened task manager and looked for it under the processes tab, but no luck. Can anyone help with this? Thank you.

If you still havn't manage to get it work or for futur OS changes,
here's a somehow easy solution :
Recreate a WinXP32bit environment on (nearly) any machine

;) (used an USB key to store all setups, iso and 7zip needed)
