I've been doing some CivV practice to try and work my way up into Deity. And I'm there. I'm late in a Deity game not dead and with the tech lead. There is hope.
But I'll probably start with the immortal equivalent because hard is fun.
I went into Civ 5 with eyes open to the fact that the game could be a buggy mess. My complaints with the game ended up being more about the design itself then bug and stability issues. I don't expect a whole Sim City issue where not only was networking not working, but the game itself was also...
I'm not going to be sitting around trying to min-max the first time through, so just based off of the backstore and lore for each faction I'll probably roll with the Peoples' African Union first and possibly shoot for a Harmony victory. That feels most appropriate.

I'm glad that I'm still undecided. I was thinking about Brasilia and going a military conquest route. But the Harmony route sounds very interesting...I think I'll kind of just go with the flow the first time win or lose.
