Altering the en_US text

How do I get the game to read the alterations I make to the en_US text file?

I've made changes so that it is English English, but nothing changes in the game.

My mistake - there was more to change than I first saw. Now have it as I want, with eg harbour instead of harbor, theatre/theater, heighbourhood/neighborhood, favour/favor etc
