I reloaded my game to find my tile with iron has disappeared. Granted, I have advanced to using saltpeter, but I think the iron is still required for knights and possibly railroads. Is this supposed to happen?

Yes - deleted happens. I think iron is the most likely of all the strategic resources to disappear. It will turn up on another spot on the map though. Try "clean map" and look for other sources.

Most of the resources will periodically disappear and reappear elsewhere. During the "tour" at the beginning of the turn where you get the little announcements about WLTK Day beginning and ending, civil disorder, quelling resisters, etc., it will show you the icon disappearing with "This source of xxxx has been depleted!" If you are lucky, the next thing you'll see is a new icon with "we have discovered a new source of xxxx!" I don't think I've ever seen it happen with horses, though....

Yes, horsies and rubber stays where they are. They're 'organic' resources and somewhat renewable.

Also, thetrooper - watch the language. :nono:

 My intention was to edit the stupidity, but I had to leave in a rush.

On topic - you will need iron for your railroads later in the game. You have salpeter for the moment: go for military tradition. Cavalry is far better than knights.

Yeah, do people find resources in general to be a lot more scarce in higher difficulty levels? They also seem to dissapear more often, and only 1/3rd of the time does a new one appear, unlike cheiftain were they will pop up 95% of the time.

Speaking of disappearing iron... I just "lost" one of my most promising AWDs to a disappeared iron.

And difficulty levels have no effect of resources.

I have to admit I'm a little annoyed at the horses - I mean, can't you just breed them?! Set up some stables in your cities and who cares about the resource tile? That aside, I love the resource system...
