When To Restart CiV

I am working on a mod right now and I am making changes that I want to check about every 10 minutes or so, so I am constantly closing an re-opening CiV. Do I have to do this every single time, or does the mod update properly while I still have CiV open?

You just have to quit back to the main menu, then go into Mods and select the mod. You'll have to enable it again if it's been updated, then start a game.
