You can save as many times as you want on your computer, to save on steamcloud you have to check the option on top of the save screen. So in your case you need to uncheck the steamcloud button.

The Alexander one means you must conquer everyone, so if you have any city states on the map you must also conquer them. The Magellan one I must have done like 9 times by now and it hasn't unlocked yet.
