We don't know yet.

If it's anything like Civ 4, the expansions will have their own 'slot'. They're marketing this as an expansion and not DLC after all.

It most likely won't seeing as the expansion achievements have already been added to the Civ V slot.

I'm also betting on a new slot as well, much like Civ IV did things. That way you can play with or without it as you like.

They have a menu option ingame (As of the expansion, we've heard) that has the option of enabling / disabling DLC, so I would presume that the expansion can be enabled / disabled as well
