If I put one of my spies in one of my cities I assume that they help keep other spies from doing spy missions on the city. If I put them in sleep mode, so I don't have to click on them every turn, do they have the same effect on other spies?

Yeah, I believe they do. Still, I don't have any proof about this :/ Just know that most of mye "Stumble upon enemy spy" has happened in cities with my own spies, or tiles with them. One thing I know for sure though is that one spy in your city can't stop every attempt from the enemy when it comes to sabotage ect.

One sleeping spy or a security bureau in a city is sufficient to improve the chance of exposing enemy spies there, but there always remains a chance of the enemy spy doing its dirty work. The same goes for a spy sleeping on your oil well. Also note that spies can only be detected when stationary, that is at the end of their move or if they haven't moved (with the intention of reducing mission cost).

Bushface is correct. After you build a Security Bureau in a city, move that city's Spy onto a critical resource or up to your border.
