Map: Pangaea Standard
Pace: Standard
Level: Emperor
Those are the most fun for me. I just got good enough to have fun on Emperor but so far I like that the AI is just strong enough to stay interesting.
Epic pace is also fun to get the most out of your units if you're more of a military minded player.

Depends upon what sort of start you want.
If you want an isolated start in the early game so you can build a few cities up before worring about the AI:
If you have Explorers: then the Tiny Islands script under advanced scripts; if not then Archepello
Large
Remove a couple of major civs and half the city states
Epic
And turn raging barbs on
And if you normally play Emperor or below, play one higher difficultly level than normal
If you want contacts but a guarantee of only two immediate neighbors along with a guartinee both your neighbors have another one: Ring map. (also requires Explorers)

I just go with continents (large), raging barbarians, and at King level.
I would also suggest that you try a mod pack (such as Hulfgars) which gives the barbarians a lot of stature. It sure makes them more challenging.

If you are new to the game I would recommend a small continents setup.
Small because it's easier to get the big picture, continents because you will have some early neighbours and late you will meet the other continent.
If you are new to TBS I would start on warlord, otherwise prince or king.
Good early builds for me are one or two scouts. On a small map I would just build one.

I would play continents or fractal map Standard size on warlord if that is to easy try prince or even king if you think you are king..
I would suggest that you play on standard speed for the first time to get know of the research agreements...
But once you have finish wih that try epic the game takes longer but it still feels like a normal game most balanced time
