Person A makes a map and plays it with Carthage.
Person B plays on the map with everything the same but with Indonesia.

???????

Then we can have actual challenges like "which civ/player can win the fastest on this map" and settle "which civ is better disputes" once and for all.

You don't need to load it into MP. You can get the seeds just by hovering over the version number at the bottom of the SP "pause" screen:

Makes this a little easier!
To borrow from my self, you can get the game and map seed using the above.
If you keep the world settings the same (eg, world age, sea level, etc.) you are free to reuse that map seed in setting up a new game. You can pick whoever you want. Unless the civ in question has a very strong start bias (like malinese for desert) there is no guarantee you'll end up in the exact same start location because it depends on what other civs are in the game. Like if you just have a river bias it might cycle between a couple options. If you keep the game seed the same too, I think you will end up with identical opponents most of the time. I've done this to truly restart a map a few times and definitely got the same map as a different civ.
