Is there any way to start a game, save it, and then load that saved game into world builder? I would like to do this with one of the larger maps from the 'Yet (not) another Maps Pack' to further customize it.

No.

Civ6 does not as yet even have a "Save map" function from the In-Game menus as Civ5 did. So far as I am aware the Civ6 Worldbuilder can still only create a new map-file or load an existing map-file (ie, X.Civ6Map type files). Save files are of extension "Civ6Save".

Not directly, but:

You can export/import a map into a mod (including from in game after loading your save) using this:
https://forums.civfanatics.com/thre...-map-using-zobtzlers-ynabmc-and-ynamp.637018/

then you can start the Worldbuilder using the map in that mod.

side note, if you want to change a map from YnAMP, be aware those are scripts, and once converted to civ6 format, you can't use anymore the YnAMP's features (not directly, I still need to write a tutorial about that)

edit: BTW you can load any of YnAMP's maps directly into the WB.

"new map" then select one as you would do to start a new game.

I know this is 6 years old, but if you are still here to read this-
I did all of the steps you outline to edit a saved game map in worldbuilder, however, my worldbuilder will not launch if YNAMP is enabled, saying "one or more mods failed to load content". any suggestions?
