How to enhance multiplayer game experience?

I've played Civ V for ages and love it but I've never actually touched the online mode before. I'm about to play a game with two friends of mine with me hosting it so I was wondering what will make it the most fun, like, should we play hotseat, pit boss etc. Thanks

Best MP games are with humans only. Join the No Quitters group and add some players that will want to play with you guys.

For hotseat/GMR games you just have to be patient of course but it's fun to deeply think about your moves it brings the game to the best level of play.

I have a weekly game with a friend of mine where we team up against the AI -- this is really a must have:

as the MP AI is extremely, extremely passive. It also allows you to use some mods (though the big ones are not supported).

We're doing a middle earth one right now with Short People Vs The World (Hobbits + Dwarves as our Team 1 vs the Humans, Elves, Goblins, and Demon-Thingees as Team 2, on Emperor, it's pretty fun -- using the middle earth civs mod).

Regular multiplayer with simultaneous turns is the way to go. Playing with all humans is much more fun if you can find reliable players who don't rage quit at the smallest set back.
