Trade route to City State

Is it because the journey to Jerusalem would require both sea and land? Can you only do one or the other?
 
Jerusalem won't have a harbor (no access to see, at least on the side where Ostia is). But there still is a chance of making a trade route to Jerusalem. If there is some other city south of Jerusalem not more than 15 tiles away from it and this city is coastal or has a harbor, you can first make a trade route to this second city. When this trade route finishes and you get the opportunity to reassign the trader, a trading post will be created in the second city. Now you can make a trade route directly to Jerusalem and the trader will use the second city as a "refueling station". It will leave Ostia as a ship, go to the second city, switch to a land trader and reach Jerusalem.

On a related subject, is religion spread via trade routes? When you set up a route, it only tells you about gold, food and production - pretty sure in Civ V it used to tell you about the religious pressure as well.
