Does a citizen need to work on a tile of a Wonder to get its yield?

Let's say a Great Library was built on Grasslands and it provides +2 Science, +1 Great Scientist point per turn

Do I get the +2 Science and +1 Great Scientist point if I don't have a citizen working on that tile?

Your problem here is you asked two different questions with the title and the post. I answered the one in the post, as written.
 
In fact, you cannot assign a citizen to work a wonder tile, however much you might try.
 
1. Wonders can not be worked, you get the benefits automatically
2. Most districts have a base adjacency yield you get automatically and then you can add up to three citizens to work the buildings as you add them for increased yields
3. Other tiles (with or without improvements) must be worked by a citizen to get any yields from that tile.
