the new mana effects, and priest promotions

No not Acheron, he is magic immune. But otherwise it works fine, I've done it on Basium.
Really? I could have sworn I'd killed acheron once with mostly fireballs. Maybe I was thinking of "hordes of sacrificial axemen" :p . I've definitely killed both Hyborem and Basium with fireballs/meteors though.

To get vaguely back on topic though, life mana's +1 health can't compete with enchantment's +1 happy either. And yeah the happy becomes fairly meaningless later on, but probably no more so than any of the other bonuses, and it's a massive legup for Grigori and Luchuirp in the very early game.

Life mana should probably give 2 health. And yeah the happy bonus sure scales up the higher the difficulty level is. On emperor its +20% Population compared to other civs at the start, but on Noble it's only 12.5% more population.

Some other of the bonuses should maybe be increased a bit. Maybe make the first mana give 10%, the 2nd 8%, the 3rd 6%, etc.

You can kill Acheron with mostly fireballs/meteors, since they do have basic, untyped strength, but it will require a lot, since he has a bonus against both fire damage and units with the summoned promotion.
