How do I take someone off my Ignore list?

Goto your User CP, then click on Edit Ignore List. Find the person you want to remove and delete their name from the list, click update list.

But you know they posted. Instead of their reply, you see a message that "You are ignoring this poster. Click <here> to read the post."
