How do you gain influence?

Check out my tourism screen - I am Spain. Definitely leading in tourism output, but I have 0/17 influence over everyone.

How do you actually go about gaining influence?

Click on the last tab "Influence by Player." That will show you how much influence you have against each opponent. You start as "Unknown" and need to get to 10% to reach "Exotic," and I think 30% gets you to "Familiar" and 60% to "Popular" (I'm at work now, so I'm going off my faulty memory). Eventually you'll get to "Influential," which is what is depicted in that screen shot.

What if I was close to Influential on one city-state? How could I go about specifically targeting to gain influence on that state? A trade route? Fulfilling its demands as they pop up? Anything I'm missing?

You're confusing Influence you get with other Civs through Tourism (which what what your original question was about) with Influence with City states (that is completely different).

In fact, the first is really called Cultural Pressure.

For City states, doing what they ask you gives influence. Also, you can click on them and use gold to buy influence.
