I know that for most starting from ancient era is the classic civ experience, and I love it too.
But I recommend trying some Later era starts once in a while.
Starting from Renaissance for example gives a certain "RFC feeling" (Rhye's and Fall Mod, coming for Civ V too!) as some of the Empires lose their early era units and some civs get more power, like America's UUs and UA become more handy.
It gets some colonial empire flavor going on too.
Medieval and Renaissance are the ones I like.
Also I like to start game from ancient, but create a turn limit, so the game stops for example on turn 200 on standard speed, and the highest score wins. Then it's more of a quickie civ fun that ends around medieval times.
I'm planning to try it on Epic or Marathon speed but I can't estimate yet how many turns I should make it last..
Anyway, I recommend trying the later starts / turn limits, it's like buying handcuffs and a John Wayne outfit to spice up marriage life, only with Civ V.
