You only get 2 turns of disease if you've got 2 citizens to lose. If it's size 2 it just drops once and it's done with for a while.
The only thing that removes disease from jungles is removing the jungles. Floodplain disease is removed by discovering sanitation; you don't actually have to build any hospitals.
