I'm a little confused about the idea of filling in specialist slots in each city. What i do currently is let a city to get to level 10+ pop and i will fill in specialists slots as long as my food stays positive and city is able to grow. This means that eventually when my city hits 30 or so pop, i have all my slots filled in for that city while still growing. My question is should i be specializing each city only for certain type of specialists, like 1 science specialist city, 1 merchants, 1 guilds? I try to build all the culture guilds in 1 city but i'm not even sure if that makes any difference or they can be spread out. Second part of this question, should i be building guilds at all, even if i'm not going for a culture victory?
 
well, yes p[ut all your guilds together if you want most benefit from other things that increase their effect.
Garden say, and ntional monument and I think some other wonders will increase great people generation.

you do want great people i think even if not going for culture win - they can defend against culture win say by giving you higher culture.

As for the original question, i would like the answer too.
Personally I fill specialist slots if a) i want the GP and b) it doesnt hurt city growth.
Sometimes when I have university i dont fill both slots if it hurts growth, but I dont know if this is optimal.
 
You can run a University specialist or two at city size 6 or 7 if you want to "start early". But it depends on if that city have the food for it or not of course.
Growth gives more science in the long run than early specialists... So its a balance act. But if you dont have the happiness for super growth, then work specialists!

Guilds should be built in the city that eventually gets the Hermitage (+50% Culture), and other %-based culture buildings/wonders.
