It was more saying to Trump "keep to your own business" in a humorous way than saying that he will seriously try to break up the US. Juncker is known to make such jokes (which is one of the reasons I think he is a great guy).
What President Juncker fails to understand though is the EU is not a sovereign nation, whereas the US is. The EU is still just an international organization akin to the United Nations. So to support secession from the EU is just showing support for the decision of a sovereign nation to not be associated with an international organization anymore, whereas supporting the secession of states from the US is actively trying to disrupt the stability of a sovereign nation and could legitimately be viewed as an act of aggression.
No, we're way past that no matter what many outside -and some still inside like the UK- believe. The UN doesn't make binding laws. The EU isn't a nation and maybe it will never been one, but the level of cooperation and integration exceeds any other alliance or treaty organization in the world and I think it will only grow now that the UK can't block anything any more.
It's a unique construct in a grey area of its own making and I see any meddling from outside forces that seek to divide us as an act of aggression, whether it's coming from Russia or the USA.

Yes. the message was very much "keep to your own business". That's not really joking matter and I doubt that Juncker is that kind of unserious person.
To what degree we're able to back it up depends how a hypothetical secession/dissolution would play out. We're certainly in no position to get involved in an American civil war should it really escalate that far. A peaceful dissolution is a different matter. Diplomatic recognition and favorable trade deals with other countries that recognize California could carry a lot of weight. We've got plenty of horses to trade and our carrot is large enough to be used as a stick.
