A Grand Brood would be the vampire nobles that rule the cities as thier own personal estates. The equivalent of Dukes or Barons which would be the governor of a given city or region.
If given the chance, they would be the planetary ruler under an emperor of an interstellar empire.
BTW that last sentence is a reference to the Dune series of books, both by the origional author Frank Herbert and his son, who now continues the legacy, Brian Herbert.

In the game, it simply gives adepts built there 1 xp per mana in the city.
Lorewise, it was used in (at least) one of the many trials Amurites go through to ascend through the ranks. The last of such trials (most Amurite mages and even archmages choose to stop long before this one) is mortal combat against the Cassawallen, in order to replace him.
I'm pretty sure that the opening bars of O Makunde is the Khazad theme. The music goes off in a wildly different place after what you hear in FFH though.
