Well, I'll probably start by checking if we are sitting on a BG. If yes, then since the settlers going to move anyway he'll start by going SE to the woods and if nothing exciting is spotted, the worker will start roading/mining the BG. On turn two the settler will create a city and make sure to work the deer. Research at max for pottery (unless there is a lot of expanionist AI) and building a warrior for scouting.
I'm not really a big fan of Spain, though I did win my first diety game (diety light) with them. The UU is quite underpowered for the era. I'm not much of a pillager, which is what that unit does best. The rules mods changing map trading, might make it more valuable when a new continent (world) is discovered.
Religious is one of my favorite traits and I plan on temples in every city. Commercial though weaker in the early game, is a strong late game trait. I'm thinking of a Bach/Smith combo for the GA. Until a little more of the map becomes unvieled, a victory condition for Spain is rather hazy, it's been a while since I've built a space ship, but I've never won a 100K game either, so both have potential. Of course there's alway the warmonger options and they both are quicker than the peaceful methods. If this is a diety game, then any victory condition is acceptable.
Our placement on the mini-map might mean a smallish continent (or even an island based) game. Map Making will be an early target unless the island ends up being larger that it appears as of now.
As for barbarians, I agree with King of America, that the barbs will be running amok.
